Web Manual - Culture of Gametophytes
Synchronization of Development In order to enhance synchronization of spore germination and early gametophyte development spores may be soaked in complete darkness for 7 days following spore sterilization, then sown as usual. Spores should only be presoaked to achieve wetting, i.e. for a few minutes, and exposure to light during the sterilization procedure should be minimized. Initial germination should occur a by two to three days following exposure to light and complete by four to six days. In addition, especially early stages of development should be more synchronous and the frequency of male gametophytes should be reduced. |
